I was thinking that maybe scattered throughout the map are /doublesize (or smaller increases in size if possible) command blocks that are one use only, so that they could serve as a sort of powerup for players to get and slowly get more powerful. It would sort of make it a deadly race, where players are trying to get to the biggest size before the others, killing the others while they do so and are distracted too.
Either that, or potions of enbiggening could work in much the same way as you just said there, as it wouldnt be too overpowered if they can get to a building and hide till it wears off for the other player. That, and two players can team up this way, one going embiggened while the other heads in the building to try and drive them out, right into the giant teamate.

Easy, I can do that with /pulse, to create a redstone block, activating a piston that blocks you from pressing the button =] I can also set the time it stays there, effectively creating a 'cooldown'
